摘要: Allele specific gene expression (ASE), with the paternal allele more expressed than maternal or vice versa, appears to be a common phenomenon in humans and mice. In other species extent of ASE is unknown, even mice there are several outstanding questions. These include; what tissue specific? how often does direction imbalance reverse between tissues? only one two alleles expressed? genome wide bias towards allele; finally do genes that nearby on chromosome share same ASE? Here we use data (RNASeq) from 18 tissues single cow investigate each these questions turn, then validate some findings 20 cows. Between 40 100 million sequence reads were generated per across three replicate samples for eighteen (the discovery dataset). A bovine atlas was created first RNASeq data), differentially identified. To analyse ASE, had access unambiguously phased genotypes all heterozygous variants cow’s whole sequence, where homozygous her sire, as result able map parental genomes, determine SNP showing tissue. total 25,251 within 7985 tested at least pervasive, 89 % significant This large proportion displaying confirmed validation dataset. For individual varied low 8–16 % those thymus high 71–82 % lung. There number cases reversed tissues. example SPTY2D1 showed almost complete kidney thymus, brain caudal lobe cerebellum. Mono allelic (MAE) common, 1349 4856 (28 %) MAE. Across tissues, 54.17 % favoured allele. Genes closely linked likely show higher (paternal maternal) expected by chance. We identified long runs neighbouring either five adjacent (GIMAP8, GIMAP7 copy1, GIMAP4, copy 2 GIMAP5) exclusive lobe. Investigating cows demonstrated 1) pervasive cattle, 2) MAE but ranges slight overexpression major allele, 3) most half time displays divergent patterns 4) 5) expressing clustered together chance, numbers
