摘要: Background and purpose: The renin-angiotensin system (RAS) regulates blood pressure electrolyte homeostasis. In addition, ‘local’ tissue-specific RAS have been identified, regulating regeneration, cell growth, apoptosis, inflammation angiogenesis. Although components of the are expressed in osteoblasts osteoclasts, a local bone has not yet described there is no information on whether involved fracture healing. Therefore, we studied expression function key component, angiotensin-converting enzyme (ACE), during healing. Experimental approach: murine femur model, animals were treated with ACE inhibitor perindopril or vehicle only. Fracture healing was analysed after 2, 5 10 weeks using X-ray, micro-CT, histomorphometry, immunohistochemistry, Western blotting biomechanical testing. Key results: hypertrophic chondrocytes periosteal callus healing, accompanied by angiotensin type-1 type-2 receptors. Perindopril treatment reduced mineral density unfractured femora. However, it improved formation, bridging gap torsional stiffness. inhibition did affect proliferation, but apoptotic death. After week treatment, smaller diameter volume indicated an advanced stage remodelling. Conclusions: Our study provides evidence for that influenced process We show first time capable accelerating remodelling.
