摘要: Polymerase chain reaction (PCR) based sequencing method called genomic amplification with direct (GAWTS) which bypasses subcloning and increases the rate of sequence acquisition by at least fivefold. The involves attachment a phage promoter onto one PCR primers. segments amplified are transcribed to further increase signal provide an abundance single-stranded template for reverse transcriptase mediated dideoxy sequencing. An end-labeled primer complementary desired generates additional specificity required generate unambiguous data.
