摘要: Abstract Rat hepatic GST 7.7 expression in cytosol and the S-hexylglutathione-agarose affinity purified fraction of from saline- (control) pyrrole-treated animals was examined using metabolic activity, SDS-PAGE, immunoblot, HPLC analyses. Metabolic activity assayed substrates 1 -chloro-2,4-dinitrobenzene, ethacrynic acid, acrolein, an ≍1.5-, 1.2-, 1.3-fold increase, respectively, monitored rate GST-catalyzed substrate conjugation to reduced glutathione. SDS-PAGE immunoblot analysis Class P1 GST-specific lgG confirmed that 7 subunit expressed fractions rats. In contrast, below limit detection saline-treated animals. demonstrated presence rat tissue showed level this increased ≍ 8-fold relative barely detectable present control tissue. N-terminal amino acid sequencing identity peak as 7-7. subunits 3, la, lb were elevated 1.7-, 2.0-, 2.4-fold, response pyrrole treatment. These data provide evidence treatment results de novo 7-7 suggest pyrrole-containing compounds potentially generated during disease and/or altered heme metabolism, or those ingested food products, may alter
