摘要: Two-photon (TP) uncaging of neurotransmitter molecules is the method choice to mimic and study subtleties neuronal communication either in intact brain or slice preparations. However, currently available caged materials are just at limit their usability have several drawbacks. The local focal nature use may for example be jeopardized by a high spontaneous hydrolysis rate commercially compounds with increased photochemical release rate. Here, using quantum chemical modelling we show mechanisms two-photon activation, synthesized more effective compounds. Furthermore, developed new enzymatic elimination removing neurotransmitters inadvertently escaping from compound during experiment. This method, usable both one experiments, allows an release. efficiency demonstrated neurophysiological experiments.
