摘要: Summary Aims This study aimed to test the diagnostic utility of total serum cell-free DNA (cfDNA) and integrity index for detection hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). Methods We initially evaluated sodium iodide (NaI) method, Triton/Heat/Phenol (THP) protocol QIAamp Kit cfDNA extraction. Then was isolated from sera 80 patients with HBV-related HCC, chronic HBV infection 50 healthy subjects, quantified by realtime quantitative polymerase chain reaction (qPCR) amplification beta-actin genomic fragments using two sets primers 100 400 bp. calculated as ratio bp β-actin fragments. Results The THP approach not only superior other methods in terms quantity, but also simpler, more rapid, less costly. Serum HCC significantly higher than that or controls. As levels, although a significant difference found between individuals, no without HCC. associated tumour size, TNM stage, lymph node distant metastasis. had sensitivity specificity discriminating DNA. Conclusions method is preferred extraction cfDNA. promising molecular biomarker detecting infection; it reflects progression metastatic potential tumour.
