Peroxisomal participation in psychosine-mediated toxicity : Implications for krabbe's disease

摘要: Psychosine (galactosylsphingosine) accumulation in globoid cell leukodystrophy (Krabbe's disease) results the loss of myelin and oligodendrocytes. To understand role psychosine toxicity Krabbe's disease, we examined effects on peroxisomal functions their relationship with reactive oxygen species. Rat C(6) glial cells were treated without cytokines. Peroxisomal beta-oxidation was significantly inhibited very long chain fatty acid levels free radicals increased cells. Furthermore, treatment decreased glutathione ATP levels, plasmalogen content, expression alkyl-DHAP synthase. Brain tissue twitcher mice (animal model Krabbe's) had activity, low glutathione, reduced plasmalogens. rat primary oligodendrocytes activities. Psychosine-mediated function radical production antioxidant N-acetylcysteine Our suggest that inhibition by may be partly responsible for oligodendrocyte observed brain, therapy useful disease.
