MRG15 is required for pre-mRNA splicing and spermatogenesis

摘要: Splicing can be epigenetically regulated and involved in cellular differentiation somatic cells, but the interplay of epigenetic factors splicing machinery during spermatogenesis remains unclear. To study these interactions vivo, we generated a germline deletion MORF-related gene on chromosome 15 (MRG15), multifunctional chromatin organizer that binds to methylated histone H3 lysine 36 (H3K36) introns transcriptionally active genes has been implicated regulation acetylation, homology-directed DNA repair, alternative cells. Conditional KO (cKO) males lacking MRG15 are sterile secondary spermatogenic arrest at round spermatid stage. There were no significant alterations meiotic division acetylation. Specific mRNA sequences disappeared from 66 germ cell-expressed absence MRG15, specific intronic retained mRNAs 4 cKO testes. In particular, encoding transition proteins replace histones sperm condensation. spermatids, colocalizes with PTBP1 PTBP2 H3K36me3 sites between exons single intron nuclear protein 2 (Tnp2). Thus, our results reveal is essential for pre-mRNA by modification could useful understand not only also, disorders underlying male infertile patients.
