摘要: This is the first report that age-associated accumulation of 8-hydroxydeoxyguanosine (8-OH-dG) does occur in human mitochondrial DNA (mtDNA) muscle diaphragm. We extracted mtDNA from diaphragm muscles differing age groups, and determined amount 8-OH-dG by ultramicro-high performance liquid chromatography / mass-spectrometry system. With same specimen, multiple deletions were detected electrophoresis after amplification polymerase chain reaction method. In subjects below 55, level was 0.02% total deoxyguanosine (dG), whereas, over 65, increased with at a rate ca. 0.25% per 10 years, reaching 0.51 % 85. Moreover, concomitant increase age. These results suggest that, younger diaphragms, replication dilutes out being not detectable. elderly aged might be slowed down leading to mtDNA, which would accelerate observed among subjects.
