摘要: Availability of tumor and non-tumor patient-derived models would promote the development more effective therapeutics for non-small cell lung cancer (NSCLC). Recently, conditionally reprogrammed cells (CRC) methodology demonstrated exceptional potential expansion epithelial from patient tissues. However, possibility to expand using CRC protocols is controversial. Here, we used approach non-tumoral biopsies patients with primary or metastatic NSCLC as well pulmonary metastases colorectal breast cancers. cultures were obtained both non-malignant tissues extraordinary high efficiency. Tumor tracked in vitro through tumorigenicity assay, monitoring tumor-specific genetic alterations marker expression. Cultures composed EpCAM+ lacking tumorigenic potential. biopsies-derived rapidly lost patient-specific mutations antigens. Similarly, colon generated cells. All examined displayed stem phenotype function. In contrast, brain failed generate cultures. conclusion, negatively selected under conditions, limiting either tissue sources. Thus, cannot be applied direct therapeutic testing cells, tumor-derived are (non-tumoral) airway basal
