摘要: Abstract α-glucosidase inhibitors (AGIs) are very attractive bioactive compounds due to their therapeutic profile that includes beneficial effects over glycemic control in type 2 diabetes mellitus and viral infections. Its detection identification plants fruits has gained growing attention, certainly requires efficient screening methodologies. The objective of the present work was develop a fast methodology detect identify AGIs cherimoya fruit (Annona cherimola Mill.) applying effect-directed analysis via high-performance thin layer-chromatography (HPTLC) linked with bioassay mass spectrometry (MS). Both, HPTLC conditions, were optimized accomplishing 50% 83% reduction on enzyme concentration incubation time respectively, compared original method. Additionally, contrast between inhibitory bands purple background also enhanced by substrate impregnation plate. conditions established following: 5.0 U mL−1 solution, 1.0 mg mL−1 2-naphthyl-α-D-glucopyranoside substrate, Fast Blue B salt solution 10 min as time. Applying this methodology, coupled HPTLC-MS ultra-high-performance liquid chromatography (UHPLC)-diode array detector (DAD)-MS/MS, it possible for first three peel seeds. Compounds tentatively assigned phenolamides (phenylethyl cinnamides): N-trans-feruloyl tyramine (m/z 314 [M+H]+; UV λmax 293 316 nm), N-trans-p-coumaroyl 284 296 nm) phenethylamine 298 288 nm). To best our knowledge, presence latter compound is reported cherimoya.
