摘要: A low nephron endowment is a strong predictor for future risk of hypertension, kidney and cardiovascular disease. Previously, we have shown that fetal uninephrectomy (uni-x) results in elevated arterial pressure, reduced renal function plasma renin activity adulthood sheep. However, the mechanisms via which reduction mass early life contributes to development hypertension disease are not well understood. Nitric oxide (NO) plays an important role driving fall vascular resistance (RVR) rise glomerular filtration rate (GFR) characterizes normal postnatal maturation newborn. We hypothesized onset dysfunction individuals with congenital due contribution NO. Fetal uni-x (n=9) or sham operation (n=7) was performed at 100d gestation (term=150 days). In conscious female lambs 6 months age, mean pressure (MAP) measured indwelling carotid catheter GFR blood flow (RBF) were clearance methods before after nitric synthase inhibition (L-NAME; 20mg/kg/h iv). Basal MAP as compared sheep (sham: 80 ± 2 mmHg, uni-x: 86 1 mmHg; P < 0.01). response systemic administration L-NAME increased similarly both groups 13 3 4 mmHg). RBF 933 29 ml/kg/h; 757 43 P<0.01) 144 107 lower decline 73 %; 47 3%; P<0.001) 54 1%; P<0.03) less Filtration fraction decreased significantly (P<0.001) but L-NAME. Renal young hypertensive born maybe underpinned by impaired bioavailability. Targeting system may offer therapeutic potential children mass, terms treatment interventions biomarker prognosis.
