Antitumor Actions of Keoxifene and Tamoxifen in the N-Nitrosomethylurea-induced Rat Mammary Carcinoma Model

摘要: We have compared the antitumor activities of antiestrogens, keoxifene (LY 156758) and tamoxifen (TAM), using N -nitrosomethylurea (NMU) rat mammary carcinoma model. To establish an effective dose TAM in this model, rats were treated 2 wk after initiation with NMU for 8 s.c. daily injections 6.25 µg, 25 or 100 µg peanut oil. At 25-µg 100-µg doses, completely inhibited tumor appearance during therapy period. An (100 daily) was to 20-, 100-, 500-µg doses 7 initiation. None keoxifene-treated groups prevented tumors as effectively 13 therapy. When at equivalent 500 starting injection, again failed prevent all 10 TAM, however, suppressed any formation. In same experiment, animals had stopped wk, started appear 6 later. No appeared when ( n = 25) continuously 23 TAM. In separate experiments, (500 administered 1 blocked binding [3H]-estradiol uteri. The effect lasted up 5 antiestrogen stopped. These experiments demonstrate that is not its action model.
