Promoter-Dependent Translation Controlled by p54nrb and hnRNPM during Myoblast Differentiation

摘要: Fibroblast growth factor 1 (FGF1) is induced during myoblast differentiation at both transcriptional and translational levels. Here, we identify hnRNPM p54nrb/NONO present in protein complexes bound to the FGF1 promoter mRNA internal ribosome entry site (IRES). Knockdown or overexpression of these proteins indicate that they cooperate activating IRES-dependent translation differentiation, a promoter-dependent manner. Importantly, transfection deletion experiments clearly demonstrate impact on activation via p54nrb hnRNPM. Accordingly, knockdown either p54 also blocks endogenous induction myotube formation, demonstrating physiological relevance this mechanism role two myogenesis. Our study demonstrates cooperative function as regulators indicates involvement mechanism.
