摘要: Enzyme-linked immunosorbent assay (ELISA) can reliably detect citrus tristeza virus (CTV) in samples collected during approximately 6 months of a typical year. Two reverse transcriptase polymerase chain reaction (RT-PCR) methods (total nucleic acid extract and immunocapture based) were evaluated compared to ELISA order develop more sensitive for CTV. From May 1994 October 1995, sweet orange trees infected with CTV from each 2 geographic areas (Riverside the San Joaquin Valley) tested monthly by method. In August (San Valley samples) September several had significant loss titer such that was not detected ELISA. By contrast, PCR gave definitive positive results these months. Different tissue types analyzed above assays. Petioles midribs, both phloem-rich tissues, satisfactory ELISA, while distal leaf tips did always produce result. All equally efficient producing result PCR-based The this study provide basis testing when drops level generally unacceptable using
