摘要: Ewing's sarcoma, the second most common malignant bone tumor, is an extremely aggressive neoplasm, mainly occurring in children and adolescents. sarcoma shows a low survival rate despite adoption of multimodal treatments, including local control disease by surgery and/or radiotherapy multidrug adjuvant chemotherapy. No new effective drugs have been recently described proposed for sarcomas and, therefore, innovative treatment modalities are very welcome needed. In this respect, two entry sites therapeutic intervention may derive from tailored therapies against insulin-like growth factor receptor I (IGF-IR) or CD99, cell surface transmembrane protein highly expressed sarcoma. Neutralizing IGF-IR functions was shown to significantly affect malignancy. However, it only that clinically applicable targeting available represent concrete opportunity. Engagement CD99 induces massive apoptosis cells through caspase-independent mechanisms reduces their potential. Since apoptotic molecule potential clinical interest, effects therapy triggering were analyzed tumors distal (lung bone) metastases athymic mice. The targeted evaluated combination with conventional chemotherapeutic agents assess best drug-drug interactions schedule. Toxic these also considered offer necessary rationale application possible forthcoming trials.
