Altered regulation of brain-derived neurotrophic factor protein in hippocampus following slice preparation.

摘要: Abstract Brain-derived neurotrophic factor (BDNF) and its cognate receptor tyrosine kinase B (TrkB) play important roles in regulating survival, structure, function of CNS neurons. One method studying the functions these molecules has utilized vitro hippocampal slice preparations. An caveat to using slices, however, is that preparation itself might alter expression BDNF, thereby confounding experimental results. To address this concern, BDNF immunoreactivity was examined rodent slices two different methods preparation. Rapid anatomically selective regulation content followed both methodologies; patterns altered were observed. First, cultured decreased dentate molecular layer increased CA3 pyramidal cell mossy fiber pathway hippocampus after 30 min. Furthermore, an initially “punctate” pattern labeling observed control sections changed homogenous . In contrast findings, prepared as for acute physiology exhibited no change min slicing, but significant increases granule layers. These findings demonstrate protein following preparation, produce regulation, raise possibility release TrkB activation may also be regulated. consequences confound analyses exogenous or endogenous on neuronal structure function.
