摘要: Recent studies on the activity of antifolates have been directed at perturbations in folate pools caused by inhibitors dihydrofolate reductase. For most part these restricted to effects methotrexate, but trimetrexate also examined. A constant feature results is elevation which accompanies inhibition both agents (1–7). variable result extent reduced coenzymes are depleted following exposure antifolates. In several cell lines reduction tetrahydrofolate quite modest (less than 40%) with 10-formyltetrahydrofolate being maintained (1–5). other cases such as L1210 cells vivo there a marked depletion coenzyme (6). Preliminary rodent hepatoma line indicated that 80% or greater and this compensated for an increase H2PteGlun 10-HCO-H2PteGlun (7). The current study extends investigations effect methotrexate human HEPG2 vitro.
