Paeoniflorin ameliorates cholestasis via regulating hepatic transporters and suppressing inflammation in ANIT-fed rats.

摘要: Paeoniflorin has shown the obvious effect on cholestasis according to our previous research. However, its mechanism not been absolutely explored yet. This study aims at evaluating potential of paeoniflorin alpha-naphthylisothiocyanate (ANIT) -induced by inhibiting nuclear factor kappa-B (NF-κB) and simultaneously regulating hepatocyte transporters. Cholestasis was induced administration ANIT. The serum indices such as total bilirubin (TBIL), direct (DBIL), aspartate aminotransferase (AST), alanine (ALT), alkaline phosphatase (ALP), γ-glutamyltranspeptidase (γ-GT), bile acid (TBA) histopathology liver were determined. Liver protein levels NF-κB, interleukin 1β (IL-1β) transporters Na+/taurocholate cotransporting polypeptide (NTCP), salt export pump (BSEP), multidrug resistance-associated 2 (MRP2) cholesterol 7α-hydroxylase (Cyp7a1) investigated western blotting. results demonstrated that could decrease ALT, AST, ALP, γ-GT, TBIL, DBIL TBA in ANIT-treated rats. Histological examination revealed rats treated with represented fewer neutrophils infiltration, edema necrosis tissue compared ANIT Moreover, significantly reduced over expressions NF-κB IL-1β tissue. In addition, relative NTCP, BSEP, MRP2 but Cyp7a1 also restored paeoniflorin. alleviating ANIT-induced seems be related reduce BSEP well MRP2.
