摘要: The trophoblast Kunitz domain proteins (TKDPs) constitute a highly expressed, placenta-specific, multigene family restricted to ruminant ungulates and characterized by C-terminal "Kunitz" domain, preceded one or more unique N-terminal domains. TKDP-1 shares an almost identical expression pattern with interferon-tau, the "maternal recognition of pregnancy protein" in ruminants. Our goal here has been determine whether ovine (ov) Tkdp-1 IFNT genes possess similar transcriptional code. ovTkdp-1 promoter cloned characterized. As promoter, is responsive Ets-2, promoter-driven reporter activity can be increased over 700-fold response over-expression Ets-2 constitutively active form protein Kinase A (PKA). Unexpectedly, element responsible for this up-regulation, unlike that IFNT, does not bind Ets-2. However, mutation CCAAT/enhancer binding within control region only reduced basal activity, but prevented as well cyclic adenosine 5'-monophosphate (cAMP)/PKA Ras/mitogen-activated kinase (MAPK) responsiveness. In vitro experiments vivo protein-protein interaction assays implicated protein-beta (C/EBP-β) involved up-regulating activity. combination C/EBP-β up-regulate minimal much 930-fold presence cAMP analog. An AP-1-like adjacent CCAAT enhancer, which binds Jun members, required cAMP/ C/EBP-β-dependent activation gene, Ets-2-dependent This paper demonstrates how key transcription factor differentiation function, two (Tkdp-1 IFNT) having spatial temporal patterns via very different mechanisms.
