摘要: Abstract AND‐34 is a member of novel family proteins (NSP1, NSP2, and NSP3) that have an amino‐terminal SH2 domain but bind by carboxy‐terminal GEF (Cdc25)‐like to the carboxy‐terminus focal adhesion adapter protein p130Cas. Direct activity toward Ras subfamily members has not been demonstrated with purified protein. Overexpression in epithelial breast cancer cells leads activation Rac Cdc42 PI3K‐dependent mechanism. This chapter will describe techniques we used examine AND‐34–induced Rac, Cdc42, Akt, PAK1 human cell lines murine lymphoid lines. In addition, summarize determine overexpression does activate R‐Ras MCF‐7 cells.
