摘要: Site-specific recombinases (SSRs) are powerful tools for genome manipulation, used in diverse organisms including Drosophila melanogaster, mouse, Arabidopsis, zebrafish, and human cultured cells. The integrase from the bacteriophage ΦC31 belongs to large serine family of integrases, contrast other widely SSRs such as Cre Flp, recombination is directional therefore irreversible. We have developed a vector system recombinase-mediated cassette exchange (RMCE) allowing swapping coding sequence an integrated transgene. Utilizing codon-optimized RNA bearing 3'UTR nanos1 gene, we replaced egfp reporter transgene with mCherry sequence. Recombination was achieved at high efficiency both somatic cells germline. demonstrate effective approach RMCE, increasing repertoire available manipulate zebrafish genome.
