The simultaneous detection of the squamous cell carcinoma antigen and cancer antigen 125 in the cervical cancer serum using nano-Ag polydopamine nanospheres in an SERS-based lateral flow immunoassay

摘要: The accurate analysis of tumor related biomarkers is extremely critical in the diagnosis early stage cervical cancer. Herein, we designed a novel and inexpensive surface-enhanced Raman scattering-based lateral flow assay (SERS-based LFA) strip with single test line, which was applied for rapid sensitive quantitative simultaneous SCCA CA125 serum samples from patients In presence target antigens, monoclonal antibody-coupled reporter-labeled nano-Ag polydopamine nanospheres (PDA@Ag-NPs) aggregated on line modified by polyclonal antibody to form double-antibody sandwich structure. finite difference time domain simulation demonstrated that large number “hot spots” generated among nanogaps PDA@AgNPs, resulted huge enhancement signal reporters. Accordingly, limit detection determined be 7.156 pg mL−1 7.182 phosphate buffer 8.093 7.370 human serum, revealing high sensitivity this SERS-based LFA strip. Significantly, using observed have specificity reproducibility, whole completed within 20 min. Furthermore, enzyme-linked immunosorbent were also employed obtained cancer, intraepithelial neoplasia healthy subjects, perfect agreement existed between both methods. Thus, clinically, developed has strong potential multiple cancer serum.
