摘要: Abstract Caveolae are flask-shaped invaginations of the plasma membrane found in many cell types, particularly endothelium. A major structural component is protein caveolin-1 which associates with numerous signalling molecules, including endothelial nitric oxide (eNOS). Caveolin-1, co-immunoprecipitates eNOS preparations from cells, regulates activity, holding it inactive. Controversy now exists regarding presence caveolae and trophoblasts, hence this study was carried out to examine whether high levels expressed human syncytiotrophoblast associated caveolin-1, find if present villous cytotrophoblasts syncytiotrophoblast. Immunohistochemistry term placentae revealed only weak labelling for although endothelium terminal villus vessels stained strongly. By electron microscopy, were identified capillary but extremely rare syncytium. Caveolin-1 staining extensive purified, isolated cytotrophoblasts, purity these cytokeratin positive cells confirmed by cytospin analysis flow cytometry. clearly demonstrated ultrastructural sections purified cytotrophoblasts. The time course expression during differentiation cytotrophoblast into culture studied. Western showed that evident day 1 whole lysates decreased at 3 when had syncytialized declined further 6, while actin (control) remained high. same samples followed a different pattern, low increasing substantially days culture, subsiding again 6. association trophoblast cultures evidenced demonstration vice versa. We conclude express assembles caveolae. Differentiation syncytium results decrease, not disappearance, marked reduction
