摘要: Abstract Protection of liver against the toxic and carcinogenic effects aflatoxin B1 (AFB1) can be achieved through induction detoxification enzymes by chemoprotectors such as phenolic antioxidant ethoxyquin. We have cloned sequenced a cDNA encoding an aldehyde reductase (AFB1-AR), which is expressed in rat response to dietary Expression Escherichia coli purification recombinant enzyme reveals that protein exhibits activity capable converting protein-binding dialdehyde form AFB1-dihydrodiol nonbinding dialcohol metabolite. show mRNA this markedly elevated rats fed ethoxyquin-containing diet, correlating with acquisition resistance AFB1. AFB1-AR represents only carcinogen-metabolizing identified date induced chemoprotector. Alignment amino acid sequence other known putative reductases shows it defines subfamily within aldo-keto superfamily.
