摘要: The stroma constitutes the structural framework of an organ and plays crucial roles in health following damage. major player with respect to extracellular matrix deposition, maintenance, remodeling is fibroblast its activated derivative, myofibroblast. It has long been recognized that there considerable variability phenotype. recent advent new single cell "omics" technologies revolutionized our understanding appreciation cellular heterogeneity fibroblasts revolutionized. With these tools, nature defining characteristics cells comprising finally being defined not just through a few markers, but by taking wholistic look at transcriptional programs. now apparent stromal are only transcriptionally diverse, also functionally, epigenetically, spatially heterogeneous. Studying populations resolution enabled identification clusters unique signatures. Whether truly represent distinct subpopulations or different states same population remains be clarified. In this chapter, we first describe procedure for purification preparation suspension from tissue samples (in case heart) RNA sequencing. We high-quality sections spatial transcriptomics. Secondly, outline workflow computational analysis sequencing transcriptomics data, as well integrating them together, explore within fibroblasts/myofibroblasts identify subtypes their locations heart.
