摘要: In present study an optimized protocol for the separation of antibodies into antigen-binding fragments F(ab')2 using pepsin digestion was investigated. The production these is a consequential step in development medical research, treatment and diagnosis. For polyclonal antibody rabbit received antigen four steps. serum at 1/128000 dilution showed high absorbance reaction with human IgG designed ELISA method. Rabbit purified by Ion-Exchange Chromatography (IEC) Purity assessed SDS-PAGE non-reduced condition only one band seen about 150 kDa MW position reduced form, two bands were 50 25 kDa positions. digested enzyme. solution applied to Gel filtration column isolate F(ab')2. Non-reduced determining purity fragment resulted 100 kDa corresponds undigested antibodies. activities FITC conjugated commercial ones compared flowcytometry activity results implied that conjugated- anti worked as efficiently one.
