摘要: Translational control plays a central role in regulation of gene expression and can lead to significant divergence between mRNA- protein-abundance. Here, we used genome-wide approaches combined with time-course analysis measure the mRNA-abundance, mRNA-translation rate protein during transition naive-to-primed mouse embryonic stem cells (ESCs). We find that ground state ESCs cultured GSK3-, MEK-inhibitors LIF (2iL) display higher ribosome density on selective set mRNAs. This mRNAs undergo strong translational buffering maintain stable levels 2iL-ESCs. Importantly, show global alteration cellular proteome pluripotency is largely accompanied by transcriptional rewiring. Thus, provide comprehensive detailed overview changes different states dissect relative contributions mRNA-transcription, translation stability controlling abundance.
