摘要: This work involves the development and validation of an enzyme immunoassay technique (EIA) for measurement cortisol concentration in cattle saliva. Saliva samples present several advantages over plasma animal welfare studies. collection avoids venipuncture as a stress factor. Also, saliva components do not affect EIA do. At present, there is no validated commercial method determination cattle. Commercially available radioimmunoassay kits human (detection range: 10-100 ng ml-1) are sensitive enough animals with low concentrations salivary (< 4 ml-1). Thus, choice Sensitivity, specificity, precision accuracy tests showed this to be suitable reliable. The detection limit was found 0.024 ml-1, representing improvement on previously described techniques. Intra-assay inter-assay variation coefficients were 1.47-7.30% 2.40-9.78%, respectively. recovery rates added 91.36-126.5%. Parallelism that levels can determined without extraction. correlation between positive (r = 0.75) saliva/plasma ratio around 10%. Therefore, alternative bovine HPA (hypothalamic-pituitary-adrenal) axis evaluation.
