Transcription ofBacillis subtilis plasmid pBD64 and expression of bacteriophage SPO1 genes cloned therein

摘要: Abstract Plasmid pBD64, a vector which is useful for cloning inBacillis subtilis ( T. J. Gryczan, A. G. Shivakumar, and D. Dubnau (1980) ,J. Bacteriol.141, 246–253), has at least three substantial transcription units. Two of these include the singleEcoRl,XbaI, andBamHI sites, while other includes singleBglII site. Each transcripts was synthesized in counter-clockwise direction, relative to pBD64 restriction map. No were detected opposite direction. Infection by bacteriophage SPO1 caused decrease each transcripts. new during SPOT infection. Various genes, cloned several tested expression observing their capacity complement mutants. Several middle late genes expressed substantially, regardless orientation fragments inserted. Since from could cause only one orientation, this argues that necessary originated promoters, and, thus, promoters can be active thymine-containing DNA.
