摘要: Reconstitution experiments with the DEAE-cellulose-treated enzymes, engaged in a two-step mechanism of synthesis alpha-glucan bound to protein, are performed. Urea/sodium dodecyl sulfate/polyacrylamide gel electrophoretic analysis radioactive products synthesized by reconstituted system shows highly glucosylated, labeled bands, whose apparent molecular masses change acrylamide concentration gels. The long carbohydrate chains during second step arise from sequential addition glucosyl moieties glucoprotein formed first step. A deglucosylation experiment confirms that product originates 38-kDa glucosylated component reaction 1 beta-amylase-sensitive moieties. Our data suggest specific phosphorylases and starch synthetases found potato tuber, which capable utilizing as primer for protein-bound glucan.
