摘要: We have investigated the physical, biochemical, and cellular properties of an autologous leukocyte platelet-rich fibrin patch. This was generated in automated device from a sample patient's blood at point care. Using microscopy, cell counting, enzyme-linked immunosorbent assay, antibody arrays, culture assays, we show that patch is three-layered membrane comprising sheet, layer platelets, leukocytes. Mean recovery platelets donated 98% (±95%CI 0.8%). levels platelet-derived growth factor AB, human transforming beta 1, vascular endothelial extracted were determined as 127 ng (±95% CI 20), 92 ng 17), 1.35 ng 0.37), respectively. showed continued release PDGF-AB over several days, rate which increased by addition chronic wound fluid. By comparison with traditional plasma, differences immune components found. The relevance these findings assessed showing mitogenic migratory effect on cultured dermal fibroblasts. Further, fibrocytes, type important for acute healing, could be grown relation to use treating recalcitrant wounds discussed.
