摘要: About 30% of the proteins encoded in genome are expressed as membrane but these represent <1% all structures solved today. In view physiological and pharmaceutical significance it is clear that a better more comprehensive understanding their three-dimensional (3D) at atomic resolution required. α-Helical integral generally difficult to work with than β-barrel-type this has particularly been true for polytopic members such large family seven-helical proteins. chapter we describe practical aspects solution-state NMR spectroscopy structure determination transmembrane (7-TM) protein receptor sensory rhodopsin pSRII from haloalkaliphilic archaeon Natronomonas pharaonis reconstituted detergent micelles. This first time 7-TM determined by NMR.
