Trinucleotide microsatellites in Norway spruce ( Picea abies): their features and the development of molecular markers.

摘要: Trinucleotide microsatellites have proven to be the markers of choice in human genetic analysis because they are easier genotype than dinucleotides. Their development can more time-consuming due their lower abundance genome. We isolated trinucleotide Norway spruce (Picea abies K.) using an enrichment procedure for genomic-library construction. Here we report on characterisation 85 ATC microsatellite-containing clones, from which 39 were developed. Many clones showed occurrence tandem repeats higher order ones, often resembling minisatellite repeats. The sequencing a sample alleles at one loci revealed size homoplasy base substitutions within microsatellite region. presence motifs repetitive sequence families was observed. found significant relationship between level polymorphism and length microsatellite. levels variability those dinucleotides, both when tested all set six individuals subset four natural populations. This difference is most likely attributable mutation rates dinucleotide loci. availability with different allows select proper marker investigate population processes time scales.
