摘要: We have tested two peptide sequences from Park9 Parkinson’s disease (PD) gene, -P 1 D 2 E 3 K 4 H 5 6 L 7 - and -F C G A N 8 9 10 (2), for Mn(II), Zn(II) Cu(II) binding. Park9 encoded protein can protect cells manganese poisoning, which is an environmental risk factor a Parkinson’s disease-like syndrome. In fact, belongs to family of ATP-ases involved in metal coordination transportation; familial gene mutations may result early development PD. The chosen fragments are located 1165 1171 1184 1193 residues the Park9 sequence, highly conserved number organisms, going yeasts humans. To understand details their binding sites at different pH values ligand molar ratios, mono- multidimensional NMR spectroscopy, together with potentiometric UV-vis experiments, been used, showing that three metals able effectively bind examined peptides. Mn(II) with involves imidazol Ne or Nδ His carboxyl γ-O Asp , Glu residues. Six donor atoms participate Mn(II) binding, resulting distorted octahedral geometry, possibly involving bidentate interaction groups; four tetracoordinated geometry. cysteine peptide; accepts additional bonds complete sphere, some water molecules.
