Effect of compounds modulating amino acid neurotransmission on the development and control of bicuculline-induced epileptogenic spiking in the rat.

摘要: Abstract Dose-related EEG spiking was induced and monitored in urethane-anaesthetised rats by cortical superfusion of bicuculline methiodide, through a cup incorporating recording electrodes. The total integrated spike voltage, number spikes as well the average size were monitored. Extracellular showed that each individual coincided with sudden, synchronous firing group superficial cells (layer II–III). γ-Aminobutyric acid reduced both frequency spikes, whilst muscimol clonazepam mainly spikes. (±) Baclofen no effect on their size. NMDA receptor antagonists, AP5 AP7 attenuating size, frequency. channel blocker MK801 also but increased at large concentrations; increasing magnesium artificial CSF, from 1 to 10 mM, had similar effect. Compounds believed preferentially block non-NMDA receptors, GAMS CNQX, activity reducing It is concluded activation GABAB receptors are important for controlling initiation bicuculline-induced GABAA control subsequent development.
