摘要: With the aim to increase number of functional markers in resource poor crop like cultivated peanut (Arachis hypogaea), large numbers available expressed sequence tags (ESTs) public databases, were employed for development novel EST derived simple repeat (SSR) markers. From 16424 unigenes, 2784 (16.95%) SSRs containing unigenes having 3373 SSR motifs identified. Of these, 2027 (72.81%) sequences annotated and 4124 gene ontology terms assigned. Among different motif-classes, tri-nucleotide repeats (33.86%) most abundant followed by di-nucleotide (27.51%) while AG/CT (20.7%) AAG/CTT (13.25%) repeat-motifs. A total 2456 EST-SSR primer pairs designed, which 366 relevance various stresses other functions, PCR validated using a set 11 diverse genotypes. 340 (92.62%) yielded clear scorable products 39 (10.66%) exhibited polymorphisms. Overall, alleles per marker ranged from 1-12 with an average 3.77 PIC 0.028 0.375 0.325. The identified EST-SSRs not only enriched existing molecular kitty, but would also facilitate targeted research marker-trait association stresses, inter-specific studies genetic diversity analysis peanut.
