DNA sequence and units of transcription of the conjugative transfer gene complex (trs) of Staphylococcus aureus plasmid pGO1.

摘要: Abstract The conjugative transfer genes of 52-kb staphylococcal R plasmid pGO1 were localized to a single BglII restriction fragment and cloned in Escherichia coli. Sequence analysis the 13,612-base region, designated trs, identified 14 intact open reading frames (ORFs), 13 which transcribed same direction. Each ORF was preceded by typical ribosomal binding sequence, 10 proteins predicted be encoded these ORFs seen when an E. coli vitro transcription-translation system used. Functional transcription units Staphylococcus aureus host complementation Tn917 inserts that abolished Northern (RNA) blot mRNA transcripts. These studies three groups (trsA through trsC, trsD trsK, trsL-trsM) four transcripts trsC [1.8 kb], trsA-trsB [1.3 trsL-trsM [1.5 trsN [400 bases]). No definite transcript for largest group, trsK (10 kb). Comparison trs-encoded amino acid sequences those data base showed 20% identity related necessary plasmids gram-negative species 32% gene required mobilization pC221 from staphylococci.
