摘要: In rice, limited efforts have been made to identify genes by the use of insertional mutagens, especially heterologous transposons such as maize Ac/Ds. We constructed Ac and gene trap Ds vectors introduced them into rice genome Agrobacterium-mediated transformation. this report, plants that contained single simple insertions T-DNA were analysed in order evaluate gene-tagging efficiency. The 3' end was examined for putative splicing donor sites. As observed maize, three splice sites identified at rice. Nearly 80% elements excised from original sites, when cDNA expressed under a CaMV 35S promoter. Repetitive ratoon culturing performed induce new transpositions derived cuttings. About 30% carried least one which underwent secondary transposition later cultures. Eight per cent transposed GUS various tissues panicles. With cloned DNA adjacent Ds, genomic complexities insertion Southern hybridization. Half showed hybridization patterns could be easily utilized locate Ds. Our data demonstrate Ac/Ds-mediated system prove an excellent tool analysis functions discuss genetic strategies employed large scale mutagenesis using Ac/Ds family
