摘要: The gamma-aminobutyric acid/benzodiazepine receptor from bovine cerebral cortex was solubilized with sodium deoxycholate and purified by affinity chromatography on benzodiazepine-agarose ion exchange chromatography. benzodiazepine binding protein enriched 1800-fold. Polyacrylamide gel electrophoresis in the presence of dodecyl sulfate dithiothreitol showed two major bands Mr = 57,000 53,000. [3H]Flunitrazepam, after UV irradiation, incorporated irreversibly into both isolated protein. A high site for acid co-purified sites were shown to reside same physical structure. dissociation constants 10 +/- 4 nM [3H] flunitrazepam 12 3 agonist [3H]muscimol. maximum specific activity muscimol 4.3 nmol/mg ratio [3H]muscimol [3H]flunitrazepam between 4. Gel filtration sucrose density gradient sedimentation studies gave a Stokes radius 7.3 0.5 nm coefficient 11.1 0.3 S, respectively. complex had pharmacological profile that corresponds specificity found membranes crude soluble extracts.
