摘要: Studies of innate immunity in fish species special concern are essential for better understanding their health status during hatchery rearing conditions. The cytochemical and morphological characterizations blood granulocytes have been used to provide information about phylogenetic differences determine the potential use neutrophil function assays. Rapid, simple, staining kits routinely mammalian characterize from four species: Arctic grayling, cutthroat trout, June sucker, shovelnose sturgeon. Blood smears were stained with Peroxidase 391 (myeloperoxidase, MPO), alkaline phosphatase (AP), Periodic Acid Schiff (PAS) Diff-quick stain; examined using bright field differential interference contrast microscopy. Granulocytes on evaluated based cell morphology, presence or absence specific chromogen. Presence lymphocytes, monocytes, platelets/thrombocytes was determined all species. trout had MPO positive granulocytes, while sturgeon heterophils reaction leukocyte AP, but not MPO. indicated measure oxidative burst degranulation primary granules sucker. Absence suggested different enzyme marker (AP) assay this Standardization techniques allowed rapid screening types, reducing number fish, time effort select adequate assays be studies concern.
