Application of high-field NMR spectroscopy for characterization and quantitation of submilligram quantities of isolated natural products

摘要: We have investigated and compared a number of sample conditions on different NMR platforms in the search maximum SNR optimal experiment time efficiency for structure elucidation quantitation natural products. Using restricted volume 3 mm Shigemi microcell assembly conjunction with 900 MHz spectrometer equipped 5 mm carbon-sensitive inverse cryoprobe, it was possible to achieve substantial increase (46-fold) as conventional room temperature 400 MHz instrument. Switching from standard tube typically improved by threefold either 600 or cryoplatform. A method that relies calibrated residual protonated solvent signal internal developed using same hardware setup tubes. Linearity spans over 3 orders magnitude, low microgram milligram quantities. successfully applied this quantify micrgram paclitaxel, verified UV/VIS measurement.
