摘要: BACKGROUND AND PURPOSE Left ventricular dysfunction following myocardial infarction is the most important predictor of adverse prognosis. Novel treatment options in require an appropriate experimental model with a standardized, hemodynamically relevant injury. We evaluated cryoinjury rodents that allows quantitative analysis systolic and diastolic dysfunction. METHODS Anesthetized, orally intubated, ventilated Lewis rats (n = 12) underwent sternotomy. Myocardial necrosis was induced by use standardized cryolesion to obtuse margin left ventricle, freezing for 3 minutes -160 degrees C. performance analyzed at day 120 after cryoinjury. Sham-operated animals 10) served as controls. RESULTS Cryoinjured behaved normally gained weight up 120. Average heart cryoinjured significantly exceeded pressure systolic, diastolic, mean aortic pressures were lower 4 months cryoinjury, whereas end-diastolic increased. had reduced blood flow, well impaired maximal dP/dt during occlusion occlusion-provoked peak pressure. Analysis rates isovolumic decrease revealed significant reduction negative animals. Finally, time constants decline prolonged CONCLUSION Standardized cryothermia induces lesion results highly reproducible impairment days cryothermia.
