Immunological identification and distribution of parathyroid hormone-like protein polypeptides in normal and malignant tissues.

摘要: Monoclonal and polyclonal antibodies recognizing human parathyroid hormone-like protein (PTHLP) have been produced using a series of recombinant synthetic PTHLP peptides. These used to develop two-site immunometric enzyme immunoassay which detects PTHLP[1-87] PTHLP[1-141] but not PTH. The detected in extracts squamous carcinomas normal tissues at concentrations from 7-515 ng PTHLP[1-87]/mg protein. Immunoblotting the extract showed highest immunoreactivity, carcinoma lung patient with hypercalcemia, revealed major band having an apparent mol wt 26,500 several other higher bands. Similar polypeptides were observed by immunoblotting cell line, SCaBER, secretes immunoreactive into its medium also tumors nude mice derived this line. Chaotropic agents did alter pattern, three different epitopes recognized these bands, indicating expression extracts. Immunohistochemical staining tissue PTHLP-containing confirmed results immunoassay, suggesting paracrine role for PTHLP. Staining was neoplastic including carcinomas, carcinoma, bladder osteogenic sarcoma, adenocarcinoma colon.
