摘要: Susceptibility to atherosclerosis is determined by both environmental and genetic factors. Its determinants have been studied use of quantitative-trait–locus (QTL) analysis. So far, 21 QTLs identified in the mouse: 7 a high-fat–diet model only, 9 sensitized (apolipoprotein E– or LDL [low-density lipoprotein] receptor–deficient mice) 5 models, suggesting that different gene sets operate each subset operates both. Among 27 human reported, 17 (63%) are located regions homologous (concordant) mouse QTLs, these same underlying genes. Therefore, genes regulating will be found most efficiently first finding their orthologs concordant QTLs. Novel QTL using combination following strategies: identifying new crosses performed with previously unused parental strains; inducing mutations large-scale, high-throughput mutagenesis screens; genomic bioinformatics tools. Once mice, they can tested association studies for relevance atherosclerotic disease.
