Effects of copper on dopaminergic function in the rat corpus striatum

摘要: Copper-loading was produced in rats by administration of 0.125% CuSO4 the drinking water for a period 11 months from weaning. At conclusion treatment animals had significant increases liver (552%) and brain (26%) copper content relative to age-matched controls. Whereas concentration dopamine unaffected, 3,4-dihydroxyphenylacetic acid corpus striatum found be lower (25% decrease) copper-treated group. Saturation studies striatal D-2 receptors using [3H]spiperone indicated that copper-loaded affinity significantly increased threefold, whereas there trend number decrease. When included radioligand binding assay, salts (Cu2+) inhibited specific untreated corpora striata. The inhibition competitive with decrease affinity, 50% effective Cu2+ 21 24 microM, potency agonists also decreased. These results are discussed relation mechanism which affects dopaminergic function use as model Wilson's disease.
