摘要: Optical imaging methods have been widely used to investigate the function of complex brain tissue. Depending on approach used, activity can be readily monitored with relatively high temporal resolution throughout large areas cortex, or alternatively from individual neurons sub-cellular compartments. The aim this chapter is illustrate application optical approaches provide information about changes in metabolism that follow coordinated neuronal firing. phenomenon spreading depression (SD) some advantages and challenges different approaches. SD involves a slowly-propagating depolarization astrocytes thought underlie migraine aura contribute enlargement injuries. stereotyped progression event across cortical surface, coupled substantial increase metabolic demand, has made response particularly amenable imaging. discussion will emphasize signals due mitochondrial redox state, as assessed by levels NADH autofluorescence. associated blood flow oxygenation also described, since these significantly impact autofluorescence studies because integration hemodynamic should valuable for understanding deficits injury accompany SD. It expected many principles discussed here apply less severe disturbances reflex activation discrete populations.
