摘要: In this study, we compared the efficiency of capture DNA probes covalently bound onto magnetic beads or microplates for their hybridization with target human cytomegalovirus (HCMV) amplicons. Polystyrene supports were first aminated by wet chemistry to allow covalent grafting probes. The level amines grafted was three times higher on than microwells. Increasingly sizes fixed both and best reaction yield ranged from 300 500 fmol. detection optimized in order obtain high yield. Long more accessible short ones target, faster kinetics obtained microplates. Sensitivity assay then determined a nonisotopic method limit found 30 amol HCMV amplicons supports. extracted clinical samples amplified PCR. resulting analyzed using sandwich discussed here. results perfectly fitted qualitative conclusions after nested PCR agarose gel.
