摘要: The pathways that contribute to thrombin-induced neuron death have been incompletely defined. Induction of cyclooxygenase 2 (COX-2), the enzyme catalyzes first step in prostaglandin synthesis, promotes neuronal injury. PGE2, a downstream product COX-2 metabolism, is neurotoxic vitro and vivo, thought be bioactive mediator responsible for neurotoxicity. objective this study determine ability thrombin affect PGE2 metabolism cultured neurons. data show apoptosis neurons, release increases when absent, regulated by dehydrogenase (PGDH), key degrades PGE2. NS398, specific inhibitor, protects neurons against toxicity, inducing active PGDH. These implicate PGDH thrombin-mediated cell death.
