摘要: An in vitro and vivo testing strategy for assessing the pulmonary effects was used to investigate safety characteristics of silica nanoparticles doped with cadmium (Cd-SiNPs). In A549 cells, Cd-SiNPs (0.5-100 μg/ml) caused (i) mitochondrial dysfunction apoptosis at 1 μg/ml, (ii) GSH depletion 10μg/ml, (iii) membrane alterations 25 after 1-day, (iv) cell growth proliferation inhibition 0.05 μg/ml prolonged exposure. Cd-SiNP were more pronounced compared CdCl2. SiNPs affected content only. results revealed early (1 day) persistent (until month) rat lung damage intratracheal instillation (1mg/rat) terms enhanced apoptotic phenomena altered parenchyma morphology. CdCl2 a delayed occurrence oxidative stress by increasing SOD1, iNOS, F2-IsoPs. The latter preceded marked increase F2-IsoPs levels plasma. did not cause stress. showed higher reactivity than SiNPs. data on toxicity suggest that is susceptible target tissue. These findings support concept multiple assays an integrated should be recommended characterize toxicological response NPs.
